Confocal microscopy, physiological, molecular, cellular, and biochemical approaches are used to study trafficking of caveolae, organization and remodeling of the cytoskeleton and cell-cell junctions, protein and fluid permeability, and signaling modules that regulate endothelial cell function. Current projects focus on 1) the signaling molecules that regulate caveolae-mediated endocytosis and transcytosis, 2) delivery of therapeutic agents across the vascular barrier, and 3) the role of caveolae in inflammation-induced increase in permeability.

Pro- inflammatory mediator thrombin induces vascular inflammation by cleaving the endothelial cell surface protease activated receptor-1 (PAR-1). PAR-1 activation causes vascular leakage and neutrophil adhesion-mediated vascular injury. Ca2+ signaling plays an important role in the mechanism of thrombin-induced vascular injury. Currently, we are investigating how thrombin increases intracellular Ca2+ by opening plasma membrane transient receptor potential channels expressed in endothelial cells. In addition, we are also investigating the role of Ca2+ signaling in the mechanism of nuclear factor ?B (NF-?B) activation- dependent expression of inflammatory genes in endothelial cells.
